Description
Testagen (Lys-Glu-Asp-Gly) is a short synthetic peptide belonging to a family of tetrapeptide bioregulators derived from tissue-specific amino acid composition studies. It has been examined in cell-culture and animal-model research for its capacity to enter the cell nucleus and interact with defined DNA sequence motifs, as well as for its activity within the pituitary-thyroid axis following hypophysectomy in avian models. The way a key is cut to fit one specific lock, Testagen and related short peptides are studied for sequence-specific binding to particular regions of genomic DNA.
Fedoreyeva et al. (Biochemistry (Moscow), 2011; PMID 22117547) documented Testagen's penetration into the nucleus of HeLa cells and its sequence-specific interaction with deoxyribooligonucleotides, noting a binding preference for CAG-containing DNA sequences. Kuznik et al. (Bulletin of Experimental Biology and Medicine, 2008; PMID 19024016) studied Testagen alongside a related tetrapeptide in neonatally hypophysectomized chickens, documenting changes in thyrotropic and thyroid hormone concentrations and thyroid gland structure over a 40-day observation period. Kuznik et al. (Advances in Gerontology, 2011; PMID 21809626) extended this hypophysectomized bird model to mature and old animals, comparing thyroid morphology and hormonal activity outcomes across age groups.
