Description
This blend combines tesamorelin, a synthetic analog of GHRH(1-44), with ipamorelin, a pentapeptide growth-hormone secretagogue studied for its selective activity at the ghrelin/GHRP receptor without engaging ACTH or cortisol pathways at the concentrations studied. The two compounds act on distinct receptors within the same axis: tesamorelin at the GHRH receptor, ipamorelin at the GHRP receptor. Much like two keys cut for adjoining locks on the same door, researchers have studied the compounds for how their distinct receptor activities intersect on pituitary somatotroph signaling.
Tomlinson reviewed tesamorelin's development as a synthetic GHRH-receptor agonist and its associated research programs (Tomlinson, Curr Opin Investig Drugs, 2006 — PMID 17086939). Falutz et al. documented increases in circulating IGF-1 associated with tesamorelin exposure in a randomized, placebo-controlled trial (Falutz et al., J Acquir Immune Defic Syndr, 2010 — PMID 20101189). Raun et al. characterized ipamorelin as the first growth-hormone secretagogue with selectivity for growth-hormone release, documenting that it did not raise ACTH or cortisol levels in animal research models even at concentrations far above its growth-hormone-release threshold (Raun et al., Eur J Endocrinol, 1998 — PMID 9849822). Venkova et al. investigated ipamorelin in a rodent research model examining gastrointestinal motility following surgery, describing its activity as a ghrelin-receptor agonist (Venkova et al., J Pharmacol Exp Ther, 2009 — PMID 19289567).
