Description
TB-500 is a synthetic version of the seven-amino-acid sequence LKKTETQ (residues 17-23), the active actin-binding site within the naturally occurring 43-amino-acid protein thymosin beta-4. The way a single gear can be removed from a larger mechanism and still turn on its own, this short fragment reproduces one specific functional site of the parent protein — the domain implicated in actin sequestration and cell motility — without carrying the rest of the molecule's structure.
Ho et al., 2012 (J Chromatogr A, PMID 23084823) characterized the N-acetylated LKKTETQ peptide as the key component of TB-500 preparations, developing an analytical method to detect the compound and its metabolites in biological samples. Philp, Goldstein & Kleinman, 2004 (Mech Ageing Dev, PMID 15037013) documented the parent protein thymosin beta-4's involvement in angiogenesis and cell migration in rodent tissue models. Sosne et al., 2010 (FASEB J, PMID 20179146) mapped the specific functional sites within thymosin beta-4, identifying the LKKTETQ actin-binding domain as distinct from the molecule's other active regions.
