Description
TB-500 is the common research name for a synthetic peptide built around the actin-binding sequence of thymosin beta-4 (Tβ4), a 43-amino-acid protein present broadly across cell types and body fluids. Structural biology has characterized how the WH2-fold motif in Tβ4 engages actin, the way a rope threaded through both ends of a pulley determines what moves and what stays fixed. The short central sequence LKKTETQ has been identified in the literature as the core active domain responsible for the protein's actin-sequestering chemistry.
Irobi et al. (2004), publishing in The EMBO Journal, resolved the crystal structure of a thymosin beta-4/actin complex, documenting how the peptide caps both ends of the actin monomer and characterizing the overlap between thymosin beta-4 and profilin binding sites. Sosne et al. (2010) mapped thymosin beta-4's biological activities to discrete short sequences within the molecule, identifying the LKKTETQ actin-binding domain as distinct from the N-terminal Ac-SDKP site. Goldstein et al. (2012) reviewed thymosin beta-4's actin-binding chemistry and its release by platelets and macrophages observed in tissue-repair research models.
