Description
SS-31 (also referenced in the literature as elamipretide) is a small, aromatic-cationic tetrapeptide designed to concentrate at the inner mitochondrial membrane by binding selectively to cardiolipin, a phospholipid unique to that membrane. The way a key fits only one specific lock among thousands, SS-31's structure allows it to bind cardiolipin with high selectivity among the many lipids present in a cell. Researchers have studied how this binding interaction affects the physical and electrochemical properties of mitochondrial membranes and the behavior of electron-transport proteins associated with them.
Mitchell et al. (J Biol Chem, 2020) used biophysical and computational methods to characterize how SS-31 partitions into lipid bilayers and alters membrane surface electrostatics, identifying this interaction as a key component of the peptide's mechanism (PMID 32273339). Birk et al. (J Am Soc Nephrol, 2013) demonstrated that SS-31 binds cardiolipin on the inner mitochondrial membrane and examined its effect on cytochrome c peroxidase activity in isolated mitochondria (PMID 23813215). Birk et al. (Br J Pharmacol, 2014) further characterized the cardiolipin-cytochrome c interaction and its relationship to electron-transport chain activity and ATP synthesis in isolated mitochondria (PMID 24134698).
