Description
SLU-PP-332 is a synthetic small molecule that engages the estrogen-related receptors (ERRα, ERRβ, and ERRγ), a family of orphan nuclear receptors implicated in mitochondrial biogenesis and skeletal-muscle metabolic signaling. Unlike naturally occurring ERR ligands, SLU-PP-332 was engineered with pharmacokinetic properties suited to use as an in vivo chemical probe. The way an accelerator pedal signals an engine to draw more fuel, SLU-PP-332 is studied for its capacity to switch on ERRα-dependent transcriptional programs associated with aerobic-exercise adaptation.
Billon et al., ACS Chemical Biology, 2023, first reported the identification of SLU-PP-332 as a pan-ERR agonist, documenting increased mitochondrial function and cellular respiration in a skeletal-muscle cell line, together with an increased oxidative muscle-fiber proportion and extended exercise endurance in a mouse model. Billon et al., Journal of Pharmacology and Experimental Therapeutics, 2024, examined SLU-PP-332 in diet-induced and genetic mouse models, documenting changes in energy expenditure, fatty-acid oxidation, and fat-mass accumulation associated with ERR activation.
