Description
Semax (ACTH(4-7)-Pro-Gly-Pro) is a laboratory-synthesized heptapeptide derived from a fragment of adrenocorticotropic hormone, extended with a proline-glycine-proline sequence that increases its stability against enzymatic breakdown. The way a locksmith copies only the notches of a key that turn the lock, Semax reproduces a narrow structural segment of ACTH associated with central-nervous-system signaling pathways, without replicating the full hormone. It is studied in rodent models for its interaction with neurotrophin gene expression and stress-signaling systems.
Stavchanskii et al. (Mol Biol (Mosk), 2011) examined Semax and its C-terminal Pro-Gly-Pro fragment for changes in neurotrophin and neurotrophin-receptor mRNA expression in rat brain tissue following carotid artery occlusion (PMID 22295573). Filippenkov et al. (Genes, 2023) used RNA-sequencing to characterize differentially expressed immune- and neurosignaling-related genes in rat frontal cortex following transient middle cerebral artery occlusion, comparing Semax with a related ACTH-derived peptide (PMID 37510287). Inozemtseva et al. (Eur J Pharmacol, 2024) studied Semax alongside a second ACTH(4-10) analog in a rodent chronic-stress paradigm, examining hippocampal BDNF levels and stress-associated behavioral markers (PMID 39442746).
