Description
Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic analog of tuftsin, an endogenous tetrapeptide, extended and modified for enzymatic stability. Enzymology research has characterized Selank as a concentration-dependent inhibitor of enkephalin-degrading enzymes in human serum, more potent in this assay than reference peptidase inhibitors such as bacitracin and puromycin. The way a decoy key jammed in a lock keeps the true key from turning, Selank is studied for how it occupies and inhibits the enzymes that would otherwise break down endogenous regulatory peptides.
Kost, Sokolov, Gabaeva, Grivennikov, Andreeva, Myasoedov, and Zozulya (Bioorganicheskaya Khimiya, 2001; PMID 11443939) demonstrated a concentration-dependent inhibitory effect of Selank on enkephalin-degrading enzymes isolated from human serum, reporting a half-maximal inhibitory concentration more potent than the reference peptidase inhibitors bacitracin and puromycin. Volkova, Shadrina, Kolomin, Andreeva, Limborska, Myasoedov, and Slominsky (Frontiers in Pharmacology, 2016; PMID 26924987) used real-time PCR to profile the expression of 84 neurotransmission-related genes in rat frontal cortex following Selank administration, documenting expression changes in a subset of genes associated with the GABAergic system within one to three hours. Kolomin, Morozova, Volkova, Shadrina, Andreeva, Slominsky, Limborska, and Myasoedov (Molecular Immunology, 2014; PMID 24291245) tracked the temporal dynamics of gene expression for a panel of signaling genes in mouse spleen tissue after Selank administration, reporting time-dependent expression changes across several markers.
