Description
PTD-DBM is a protein transduction domain fused to the Dishevelled-binding motif of CXXC5, a zinc-finger protein identified as a negative feedback regulator of Wnt/β-catenin signaling. The way a key can be cut to block a specific lock without touching any other lock in the building, PTD-DBM is designed to interrupt one specific protein-protein interaction — CXXC5 binding to Dishevelled — while leaving the rest of the signaling network untouched.
Lee et al., 2015 (J Exp Med, PMID 26056233) first characterized PTD-DBM in a mouse model of cutaneous wound repair, documenting activation of β-catenin and collagen production when the CXXC5–Dishevelled interaction was disrupted. Ryu et al., 2023 (Cells, PMID 36831222) investigated PTD-DBM in a wound-induced hair neogenesis model, examining the CXXC5-mediated pathway alongside prostaglandin D2 signaling. Lee et al., 2023 (Advanced Healthcare Materials, PMID 36854308) studied PTD-DBM delivered via a hydrogel patch system in a mouse wound model, examining downstream markers of tissue regeneration.
