Description
PNC27 is a synthetic peptide containing a segment of the p53 transactivation domain (residues 12-26) joined to a membrane-penetrating sequence. Membrane-bound HDM-2 has been documented on the surface of certain transformed cell lines but not on untransformed cells studied alongside them. Much like a key that fits only one lock, PNC27 has been studied for its selective binding to this membrane-bound HDM-2, forming transmembrane pores in cells that express it while leaving HDM-2-negative cells in the same culture undisturbed.
Krzesaj et al. (2024), published in Annals of Clinical and Laboratory Science, documented PNC27 binding to the p53-binding site of membrane HDM-2 using a monoclonal-antibody blocking assay, and observed mitochondrial membrane disruption in treated pancreatic carcinoma cell lines via immuno-electron microscopy. Pincus et al. (2024), in Biomedicines, characterized the pore-formation mechanism, describing PNC27's complex formation with HDM-2 followed by dimerization into transmembrane channels across a range of cell-line models studied. Krzesaj et al. (2025) extended this line of research to a cervical squamous cell line, documenting HDM-2 membrane expression as the determining factor for peptide-associated cytotoxicity observed in vitro.
