Description
PE 22-28 is a synthetic, shortened analog of spadin, a peptide identified as a proteolytic fragment of the protein sortilin. Spadin and its shortened analogs have been studied for their affinity toward TREK-1 (TWIK-related potassium channel-1), a two-pore-domain potassium channel implicated in neuronal excitability. The way a lock accepts only a matching key, PE 22-28 is studied for a tighter, more selective binding affinity to the TREK-1 channel than its parent peptide, spadin.
Djillani et al. designed PE 22-28 from spadin blood-degradation products and reported, using patch-clamp electrophysiology on hTREK-1/HEK cells, a TREK-1 channel IC50 of 0.12 nM for PE 22-28 compared to 40-60 nM for spadin; in mouse behavioral assays including the forced swim test and novelty-suppressed feeding test, PE 22-28 was associated with altered immobility-time and feeding-latency measures, along with markers of hippocampal neurogenesis and increased PSD-95 expression in cortical neurons (Front Pharmacol, 2017, PMID 28955242). Spadin, the parent peptide, has been studied for selective TREK-1 blockade without the pain-, seizure-, or cardiac-conduction changes associated with other TREK-1-targeting approaches in rodent models (Moha Ou Maati et al., Neuropharmacology, 2012, PMID 21807005).
