Description
Orexin-B is the second of two neuropeptides generated by proteolytic processing of the prepro-orexin precursor. It shares partial sequence identity with orexin-A but shows distinct selectivity for the OX2 receptor subtype. Alongside orexin-A, it was identified as the endogenous ligand for a pair of previously orphan G protein-coupled receptors located in the lateral hypothalamus, and it continues to be used as a research tool for isolating OX2-specific signaling from the broader orexin system.
Sakurai et al. (1998, Cell) identified orexin-B alongside orexin-A as products of the same precursor peptide and characterized their binding activity at two closely related G protein-coupled receptors. Dale et al. (2026, Biochemical Pharmacology) compared the intracellular signaling kinetics of OX2 receptor activation by orexin-A versus orexin-B using bioluminescence resonance energy transfer assays, reporting ligand-dependent differences in G-protein trafficking and beta-arrestin recruitment. Prischich et al. (2024, Cellular and Molecular Life Sciences) developed a photoswitchable analog of orexin-B for in vitro and in vivo receptor-pharmacology studies, demonstrating light-dependent control of OX2 receptor activity in zebrafish larvae.
