Description
Orexin-A is one of two neuropeptides, alongside orexin-B, derived from a common prepro-orexin precursor by proteolytic processing. It was first localized to neurons within the lateral and posterior hypothalamus and identified as the endogenous ligand for two closely related G protein-coupled receptors, OX1 and OX2, that had previously been classified as orphan receptors. The way a pilot light keeps a larger heating system primed and ready, orexin-A-expressing neurons are studied for their role in maintaining baseline arousal and metabolic signaling within the hypothalamic circuits they innervate.
Sakurai et al. (1998, Cell) identified orexin-A and its receptors, reporting that central delivery of the peptide to rats was associated with increased food consumption and that prepro-orexin mRNA expression was upregulated during fasting in the animal model. Olsen, Furlong, and Carrive (2021, Behavioural Brain Research) examined microinjection of orexin-A into the central amygdala of rats, documenting changes in heart rate, arterial pressure, and general activity relative to vehicle controls. Pan et al. (2024, British Journal of Pharmacology) investigated orexin-A delivery in rodent and feline motion-sickness models (intracerebroventricular administration in rats; intranasal administration in cats), reporting behavioral effects linked to OX1 and OX2 receptor activity across several hypothalamic nuclei.
