Description
Neuropeptide Y (NPY) is a 36-residue peptide amide, structurally related to peptide YY and pancreatic polypeptide, first isolated and sequenced from porcine brain tissue. It is among the most abundant peptides in the mammalian central nervous system, distributed across hypothalamic, limbic, and autonomic signaling circuits. The way a thermostat's setpoint is read by multiple downstream systems at once, NPY is studied as a signal read by several distinct receptor subtypes (Y1, Y2, Y5, among others), each linked to a different arm of hypothalamic, endocrine, and limbic research.
Tatemoto (1982, Proceedings of the National Academy of Sciences) first determined the complete amino acid sequence of NPY from porcine brain, establishing its structural homology to peptide YY and pancreatic polypeptide and defining it as a founding member of this peptide family. Clark et al. (1987, Regulatory Peptides) administered NPY and related analogs directly into the third ventricle of rats, documenting concentration-dependent increases in food-intake behavior and time spent eating relative to vehicle controls. Heilig and Widerlöv (1995, Critical Reviews in Neurobiology) reviewed the neuroanatomy and physiology of central NPY systems, summarizing research on its involvement in hypothalamic-pituitary signaling, circadian rhythm, and limbic circuits, and describing NPY's characterization in animal models as an endogenous anxiolytic-like signal within that research literature.
