Description
Mechano growth factor (MGF) is the common name for the IGF-1Ec splice variant, a peptide sequence encoded by an alternate reading of the insulin-like growth factor-1 (IGF-1) gene. Unlike mature IGF-1, MGF carries a distinct E-domain sequence that has been investigated in cell-culture models for signaling activity separate from the primary IGF-1 receptor pathway. Research models have examined MGF expression as a mechanically induced, localized signal in skeletal muscle tissue.
Yang and Goldspink, 2002 investigated the distinct E-domain of MGF (IGF-1Ec) in myoblast cell culture, finding that its signaling activity on proliferation and differentiation differed from that of mature IGF-1 and was not fully blocked by an IGF-1 receptor antibody (FEBS Lett, PMID 12095637). Owino et al., 2001 examined MGF expression in rat skeletal muscle following mechanical overload, documenting reduced local MGF expression in older animals compared to younger ones (FEBS Lett, PMID 11566187). Cheema et al., 2005 studied IGF-1 gene splicing in a three-dimensional muscle-cell culture model, observing that MGF and IGF-1Ea expression were both induced by mechanical strain, with cyclical-loading regimens differentiating the two splice variants (J Cell Physiol, PMID 15389530).
