Matrixyl (palmitoyl pentapeptide-4, chemical designation C16-KTTKS) is a synthetic lipopeptide formed by attaching a 16-carbon palmitoyl chain to the pentapeptide sequence Lysine-Threonine-Threonine-Lysine-Serine. The way soap molecules arrange themselves into organized structures in water, C16-KTTKS self-assembles into fibrillar tape-like nanostructures with high beta-sheet content, a structural property studied alongside the peptide's documented activity in fibroblast cell-culture research.
Jones, Castelletto, Connon, and Hamley (Mol Pharm 2013; PMID 23320752) examined C16-KTTKS in contact with human dermal and corneal fibroblast cultures, reporting a concentration-dependent stimulation of collagen production that tracked closely with the compound's critical aggregation concentration, suggesting a link between the peptide's self-assembly behavior and its collagen-related activity. Castelletto, Hamley, Whitehouse, Matts, Osborne, and Baker (Langmuir 2013; PMID 23822110) used spectroscopic, microscopic, and X-ray scattering methods to characterize C16-KTTKS self-assembly, finding that the compound forms flat tapes and extended fibrillar structures with high beta-sheet content and a cross-beta X-ray diffraction pattern, distinguishing it structurally from related palmitoylated peptides examined in the same study. Tałałaj, Uścinowicz, Bruzgo, Surażyński, Zaręba, and Markowska (Molecules 2019; PMID 31618846) synthesized a series of novel KTTKS analogues and tested their effects on fibroblast cytotoxicity and collagen and DNA biosynthesis, reporting that none of the synthesized peptides were cytotoxic to fibroblasts at the concentrations tested.
