Description
Liraglutide is a 31-amino-acid peptide analog of human glucagon-like peptide-1, modified with a 16-carbon fatty acid side chain attached through a glutamic acid spacer. The way a heavier anchor holds a boat in place longer against a moving current, this fatty-acid modification is associated with binding to circulating plasma proteins, a structural design investigated for extending the peptide's presence in plasma relative to unmodified glucagon-like peptide-1.
Madsen, Knudsen, Agersoe, Nielsen, Thøgersen, Wilken, and Johansen (J Med Chem 2007; PMID 17975905) reported a structure-activity series describing how fatty acid chain length, polarity, and bulkiness govern receptor potency and plasma protraction of glucagon-like peptide-1 fatty acid derivatives, including liraglutide, identifying fatty acid length as the parameter most closely tied to protraction. Plum, Jensen, and Kristensen (J Pharm Sci 2013; PMID 23853127) developed a reiterated stepwise equilibrium dialysis assay to quantify liraglutide's plasma protein binding, reporting greater than 98.9% binding at a clinically relevant concentration, with specific binding to human serum albumin measured at 99.4%. Pyke, Heller, Kirk, Ørskov, Reedtz-Runge, Kaastrup, Hvelplund, Bardram, Calatayud, and Knudsen (Endocrinology 2014; PMID 24467746) used a validated monoclonal antibody to map glucagon-like peptide-1 receptor localization across monkey and human tissue by immunohistochemistry, identifying receptor expression in pancreatic beta cells, vascular smooth muscle, cardiac sinoatrial node myocytes, and specific gastrointestinal cell populations.
