Description
Des(1-3)IGF-1 is a naturally occurring truncated variant of IGF-1 missing the N-terminal tripeptide Gly-Pro-Glu. The absence of this fragment changes its interaction with IGF-binding proteins in ways researchers have characterized extensively in cell and animal models. The way a key removed from a lock changes which doors it opens, removing this tripeptide is thought to change which binding partners the molecule can engage in cellular signaling pathways.
Ballard et al. characterized Des(1-3)IGF-1 as a naturally occurring truncated variant with markedly reduced binding to IGF-binding proteins compared to native IGF-1, a property linked to greater potency in cultured-cell proliferation assays (Ballard et al., Int J Biochem Cell Biol, 1996 — PMID 8930132). Tomas et al. compared Des(1-3)IGF-1 against native IGF-1 and related analogs in pig and marmoset research models, examining hypoglycaemic potency and plasma glucose response across the analog series (Tomas et al., J Endocrinol, 1997 — PMID 9415072). Walton et al. reviewed a series of pig studies characterizing how variants with weaker IGF-binding-protein affinity differ from native IGF-1 across growth and metabolic research endpoints (Walton et al., Prog Growth Factor Res, 1995 — PMID 8817682).
