Description
Humanin is a 24-amino-acid peptide first identified through a functional screen of Alzheimer's disease brain cDNA, encoded within a region of the mitochondrial genome overlapping the 16S ribosomal RNA sequence. The way a cell's mitochondria send distress signals to the rest of the cell, Humanin has been characterized in the literature as a secreted signaling peptide studied across cellular stress-response and metabolic-signaling research pathways.
Tajima, Niikura, Hashimoto, Ito, Kita, Terashita, Yamazaki, Koto, Aiso, and Nishimoto (Neurosci Lett 2002; PMID 12009529) raised an antibody against Humanin and detected the native peptide in mouse testis and colon tissue as well as in human brain tissue samples, establishing that the peptide is produced in vivo rather than existing only as a theoretical open reading frame. Takeshita, Hashimoto, Nawa, Uchino, and Matsuoka (J Biol Chem 2013; PMID 23861391) investigated the intracellular signaling pathway downstream of Humanin receptor binding in neuronal cell culture, identifying SH3 domain-binding protein 5 as a transcriptional target that binds and inhibits c-Jun N-terminal kinase. Muzumdar, Huffman, Atzmon, Buettner, Cobb, Fishman, Budagov, Cui, Einstein, Poduval, Hwang, Barzilai, and Cohen (PLoS One 2009; PMID 19623253) examined central and peripheral infusion of Humanin and Humanin-derived analogs in rodent models, reporting activation of hypothalamic STAT-3 signaling and an age-associated decline in circulating Humanin levels in both rodents and humans.
