Description
Glutathione (GSH) is a tripeptide composed of glutamate, cysteine, and glycine, synthesized endogenously in the cytosol of mammalian cells. It has been characterized in the literature as a ubiquitous intracellular thiol with roles in detoxification, redox buffering, and cell-proliferation signaling. The way a rechargeable battery cycles between charged and depleted states, glutathione is described in research as cycling between reduced (GSH) and oxidized (GSSG) forms as part of cellular oxidative-stress buffering.
Lu (Molecular Aspects of Medicine, 2009; PMID 18601945) reviewed the enzymatic regulation of glutathione synthesis, identifying glutamate-cysteine ligase as the rate-limiting enzyme and cysteine availability as a primary determinant of cellular glutathione capacity. Richie et al. (European Journal of Nutrition, 2015; PMID 24791752) conducted a six-month randomized, placebo-controlled trial in healthy adults and documented elevated blood, erythrocyte, plasma, and buccal-cell glutathione levels following sustained oral intake, alongside a shift in the oxidized-to-reduced glutathione ratio. Sinha et al. (European Journal of Clinical Nutrition, 2018; PMID 28853742) examined a liposomal oral glutathione formulation in a pilot study and documented parallel changes in natural killer cell cytotoxicity and lymphocyte proliferation markers alongside glutathione level increases.
