Description
FST 344 refers to an alternatively spliced isoform of the activin-binding protein follistatin, encoded by a 315-amino-acid secreted variant that lacks the heparin-binding domain present in full-length follistatin. Because it circulates systemically rather than binding cell-surface sites, this isoform has been studied in muscle-biology and gene-therapy research as a way to antagonize the TGF-β superfamily member myostatin (GDF-8) while limiting off-target activity at the hypothalamic-pituitary-gonadal axis associated with full-length follistatin.
Rodino-Klapac et al. (2009, Muscle & Nerve) reviewed the FS344 splice variant as a strategy for avoiding off-target effects associated with full-length follistatin, describing translational studies spanning mice to nonhuman primates. Kota et al. (2009, Science Translational Medicine) documented that AAV1-delivered FS344, targeted to skeletal muscle, was associated with durable increases in muscle size and strength in cynomolgus macaques, with no organ-morphology abnormalities observed over the study period. Reichel et al. (2019, Drug Testing and Analysis) developed an electrophoretic detection method for unauthorized "follistatin 344" preparations circulating outside regulated research channels, noting its listing under the WADA Prohibited List.
