Description
FOXO4-DRI is a D-retro-inverso isoform of a FOXO4-derived peptide, an amino-acid-reversed, D-configured design intended to confer protease resistance relative to the parent L-peptide. Research models have examined the compound's disruption of the FOXO4-p53 interaction and the resulting nuclear exclusion of p53 in senescent cells. The way a mirror-image key can be cut to resist a standard lock-picking approach, D-retro-inverso peptides like FOXO4-DRI are studied for extended stability against enzymatic breakdown compared with their L-amino-acid counterparts.
Kong, Li, Liu, and colleagues (Commun Biol 2025; PMID 39994346) investigated FOXO4-DRI in keloid fibroblast and organ-culture models, examining its association with p53-serine-15 nuclear exclusion and apoptosis in senescent fibroblast populations. Li, Zhang, Cheng, and colleagues (Exp Gerontol 2024; PMID 39025385) studied FOXO4-DRI in a model of Leydig cell senescence in aged mice, examining apoptosis induction in senescent cells and secretory phenotype markers in cocultured cell systems. Liu, Hou, Wang, Liu, and Cheng (Naunyn Schmiedebergs Arch Pharmacol 2023; PMID 37074394) examined FOXO4-DRI in a bleomycin-induced pulmonary fibrosis mouse model, studying its association with extracellular matrix protein content and p53 nuclear distribution in fibroblasts.
