Description
FOXO4 is a member of the forkhead box class O (FOXO) family of transcription factors. Structural biology research has examined how the FOXO4 forkhead DNA-binding domain engages the p53 transactivation domain, an interaction studied in the context of senescent cell viability. The way removing a single wedge can release a jammed mechanism, a peptide designed to perturb the FOXO4-p53 interaction is studied for its selective effect on senescent cell populations.
Baar, Brandt, Putavet, and colleagues (Cell 2017; PMID 28340339) identified FOXO4 as a factor in senescent cell viability and designed a FOXO4-derived peptide to perturb the FOXO4-p53 interaction, studying its effects on cell-intrinsic apoptosis in senescent cell populations and in mouse models. Kohoutova, Srb, Obsilova, Veverka, and Obsil (Nat Commun 2025; PMID 40425537) used NMR and structural biology methods to characterize the heterogeneous binding modes between the FOXO4 forkhead domain and the p53 transactivation domain. Asadi, Moundounga, Chakroborty, Pokokiri, and Wang (Front Mol Biosci 2025; PMID 40260403) reviewed the FOXO family of transcription factors, including FOXO4, and their regulation by the PI3K-Akt pathway in cell survival and stress-response research.
