Dermorphin is a seven-amino-acid, D-amino-acid-containing peptide first isolated from Phyllomedusa frog skin secretions and characterized as a heptapeptide with potent opiate-like activity (Montecucchi et al., Int J Pept Protein Res, 1981; PMID 7287299). Description
Dermorphin is a seven-amino-acid peptide, notable in peptide chemistry for containing a D-amino acid residue — a structural feature rarely found in vertebrate-derived peptides. First isolated from methanol extracts of Phyllomedusa sauvagei skin, dermorphin has been characterized in receptor-binding and functional laboratory assays as a high-affinity ligand for the mu-opioid receptor. The way a single key can be cut to fit one specific lock exceptionally well, dermorphin's D-amino acid-containing sequence is described in the literature as conferring unusually high receptor selectivity relative to endogenous enkephalins.
Montecucchi et al. (International Journal of Peptide and Protein Research, 1981; PMID 7287299) first identified dermorphin's amino acid sequence and reported its potent opiate-like activity in the originating isolation study. Bird et al. (PLoS One, 2016; PMID 27272042) characterized a bivalent Mu-NOP ligand built from dermorphin and the nociceptin/orphanin FQ peptide across a panel of receptor-binding and functional laboratory assays. Steel et al. (Drug Testing and Analysis, 2014; PMID 24259424) developed a liquid chromatography-tandem mass spectrometry screening method for seventeen dermorphin-class peptides in equine and human biological matrices, a method developed for anti-doping laboratory settings.
