Description
CJC-1295 without DAC is a synthetic, tetra-substituted analog of the growth-hormone-releasing hormone fragment GRF(1-29), distinguished from the DAC-conjugated form of CJC-1295 by the absence of the fatty-acid linker that binds circulating albumin. Without that modification, the peptide's structure more closely resembles the native GRF(1-29) backbone. Amino-acid substitutions across this backbone class have been mapped in receptor-binding studies — the way a locksmith tests which notches on a key blank can be altered without changing whether it still turns the lock — to identify which positions tolerate substitution without disrupting receptor affinity.
Lefrançois and Gaudreau (Neuroendocrinology, 1994) mapped receptor-binding affinity across a systematic series of amino-acid-substituted GRF(1-29) analogs in rat adenopituitary tissue, identifying which positions in the peptide backbone tolerate substitution without loss of receptor affinity (PMID 8202217). Cristea et al. (Anal Biochem, 2023) developed and validated a UHPLC-MS/MS analytical method for quantifying CJC-1295 and related growth-hormone-releasing hormone peptide analogs (PMID 37806509). Uçaktürk and Nemutlu (J Pharm Biomed Anal, 2026) developed a complementary nano-LC-Orbitrap mass spectrometry method for the analytical characterization of CJC-1295 and other growth-hormone-releasing hormone analogs (PMID 41138283).
