Description
CAG is a long-acting analogue of amylin, a pancreatic peptide hormone, chemically modified with a fatty-acid moiety for extended plasma exposure. It has been investigated alongside a second synthetic peptide sequence, referred to here by internal designation Bio-1s, across early-phase clinical research examining pharmacokinetics, tolerability, and safety when the two compounds are studied together. The way two instruments recorded on separate tracks can be studied individually and as a mix, CAG and Bio-1s have been evaluated both as standalone research subjects and as a co-administered pair in the cited literature.
Lau et al. (Lancet, 2021; PMID 34798060) conducted a multicentre, randomized, placebo-controlled and active-controlled phase 2 trial of CAG alone across ten countries, examining a range of CAG doses and measuring safety, tolerability, and pharmacodynamic endpoints in adult study participants. Enebo et al. (Lancet, 2021; PMID 33894838) conducted a phase 1b trial evaluating the concomitant administration of CAG with Bio-1s, measuring pharmacokinetic and pharmacodynamic parameters — including exposure, half-life, and clearance — of both compounds when studied together. Lau et al. (Journal of Medicinal Chemistry, 2015; PMID 26308095) documented the chemical design process for the Bio-1s peptide sequence, describing the fatty-acid conjugation strategy used to extend plasma half-life relative to earlier peptide analogues, characterized in mini-pig pharmacokinetic studies.
