Description
B7-33 is a single-chain derivative of the B-chain of human gene-2 (H2) relaxin, designed to bind the relaxin family peptide receptor 1 (RXFP1) while biasing downstream signaling toward the ERK1/2 pathway over the cAMP pathway used by native two-chain relaxin. It has been characterized in the literature as the first functionally selective RXFP1 agonist, distinguishing its receptor-signaling profile from that of full-length H2 relaxin. The way a key cut to trip only one tumbler in a lock can still open the door, B7-33 is described in research as activating a narrower slice of RXFP1 signaling than its parent hormone.
Hossain et al. (Chemical Science, 2016; PMID 30155023) designed and characterized B7-33 as a single-chain RXFP1 agonist biased toward ERK1/2 signaling, and documented its activity across three preclinical rodent models of heart and lung fibrosis. Alam et al. (Biomedicine & Pharmacotherapy, 2023; PMID 36753958) compared B7-33 to full-length relaxin and to the ACE inhibitor perindopril in an isoprenaline-induced cardiomyopathy mouse model, reporting left ventricular fibrosis, inflammation, and vascular measures across treatment groups. Abboud et al. (Pain Reports, 2021; PMID 34159282) examined central RXFP1 activation by B7-33 and H2-relaxin delivered intracerebroventricularly in a mouse model of inflammatory pain, reporting changes in mechanical and thermal sensitivity alongside RXFP1 expression mapping in pain-processing brain regions.
