Description
Alprostadil is the synthetic form of prostaglandin E1 (PGE1), a lipid signaling molecule derived from arachidonic acid. It has been documented in primary literature as an agonist at prostaglandin E-series receptors coupled to adenylate cyclase, elevating intracellular cyclic AMP (cAMP) in smooth muscle tissue. The way a dimmer switch adjusts current to change a room's lighting, cAMP accumulation is thought to shift the contractile state of smooth muscle cells toward relaxation.
Fullerton et al. (J Surg Res, 1994; PMID 7913147) documented that PGE1 relaxed isolated rat pulmonary arterial rings via a cAMP-mediated, prostaglandin-receptor pathway, comparing its potency to cGMP-mediated and beta-adrenergic relaxation pathways in the same tissue model. Fullerton et al. (J Surg Res, 1996; PMID 8656622) further characterized PGE1 as an EP2-receptor agonist among several cAMP-linked pathways studied in isolated pulmonary artery rings, noting differences in potency and efficacy across receptor systems. Palmer et al. (J Urol, 1994; PMID 8072124) documented concentration-dependent cAMP accumulation in cultured human smooth muscle cells following PGE1 exposure, and found this cAMP response correlated with the magnitude of smooth muscle relaxation observed in isolated tissue strips.
