Description
ACE-031 is a fusion protein combining the extracellular domain of human activin receptor type IIB (ActRIIB) with the Fc region of human IgG1. It is engineered to bind myostatin and related TGF-β superfamily ligands, the signaling proteins that constrain skeletal muscle growth. Research on ACE-031 spans single-administration pharmacology studies, a randomized controlled trial, and non-human primate models examining its effects on muscle mass, strength, and body composition.
Attie et al. (2013) conducted a double-blind, placebo-controlled ascending-dose study of a single administration of ACE-031 in healthy adult volunteers, measuring total body lean mass and thigh muscle volume to characterize the pharmacodynamics of activin-receptor pathway modulation. Campbell et al. (2017) documented a randomized, double-blind, placebo-controlled trial of ACE-031 administered subcutaneously in ambulatory boys with Duchenne muscular dystrophy, examining safety, pharmacokinetics, and pharmacodynamic endpoints including lean body mass, fat mass, bone mineral density, and the six-minute walk test; the trial was discontinued after non-muscle-related adverse events (epistaxis and telangiectasias) were observed. Cadena et al. (2026) examined ACE-031 in the common marmoset, a non-human primate model, measuring lean body mass and muscle fiber cross-sectional area over a 14-week study period.
