5-Amino-1MQ (5-amino-1-methylquinolinium) is a synthetic small molecule belonging to the methylquinolinium chemical class, characterized in the literature as a selective, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT). NNMT is a cytosolic enzyme that transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, a reaction implicated in cellular NAD+ and SAM homeostasis. The way throttling one valve in a plumbing system changes pressure throughout the whole network, inhibiting NNMT is described in the research literature as altering the balance of several interconnected metabolic cofactor pathways.
Neelakantan et al. (Biochemical Pharmacology, 2018; PMID 29155147) characterized methylquinolinium-scaffold NNMT inhibitors, documenting enzyme selectivity against related methyltransferases and changes in adipocyte lipogenesis and intracellular NAD+/SAM levels in cultured cells and diet-induced obese mice. Dimet-Wiley et al. (Scientific Reports, 2022; PMID 35013352) treated diet-induced obese mice with 5-amino-1-methylquinolinium in combination with a reduced-calorie diet and documented changes in whole-body adiposity, body weight measures, and gut microbiome composition relative to diet-switch-alone and lean control groups. A related small-molecule NNMT inhibitor from the same chemical series was investigated by Neelakantan et al. (Biochemical Pharmacology, 2019; PMID 30753815) in aged skeletal muscle research models, where treated mice showed elevated muscle stem cell proliferation and fusion following induced injury.
