Description
Tesamorelin is a synthetic peptide analog of human GHRH(1-44), engineered for greater stability than the native hormone. As a GHRH-receptor agonist, it is studied for its role in the pituitary somatotroph signaling axis — the same axis responsible for growth-hormone secretion. The way a key is cut to fit a specific lock, tesamorelin's structure is designed to engage the GHRH receptor with the specificity the native hormone shows.
Tomlinson reviewed tesamorelin's pharmacology as a synthetic GHRH analog, documenting its development as a GHRH-receptor agonist and the clinical research programs examining its activity across several research contexts (Tomlinson, Curr Opin Investig Drugs, 2006 — PMID 17086939). Falutz et al. conducted a randomized, placebo-controlled trial in adults that documented increases in circulating insulin-like growth factor-1 (IGF-1) associated with tesamorelin exposure over a six-month research period, a marker of downstream GHRH-receptor and pituitary-axis activity (Falutz et al., J Acquir Immune Defic Syndr, 2010 — PMID 20101189). A subsequent 52-week extension of that trial documented the compound's tolerability profile over sustained exposure (Falutz et al., AIDS, 2008 — PMID 18690162).
