Description
Mazdutide is a synthetic peptide analog derived from gut-hormone signaling sequences. It has been investigated in preclinical and clinical research examining hepatic lipid handling and inflammatory signaling pathways in cellular and rodent models, as well as in clinical trial data compiled across multiple research cohorts.
Kamrul-Hasan et al. (Diabetes Obes Metab, 2026; PMID 42410325) conducted a systematic review and meta-analysis of randomized controlled trials examining mazdutide — a dual GLP-1/glucagon receptor agonist — versus placebo and active comparators in adults with overweight/obesity and/or type 2 diabetes; the analysis pooled endpoints including body weight, HbA1c, waist circumference, lipid profile, liver enzymes, and safety across nine trials. Gan et al. (Pharmaceuticals, 2026; PMID 41901218) investigated mazdutide in a high-fat-diet mouse model and a free-fatty-acid-treated hepatocyte model, examining the endoplasmic-reticulum-stress PERK pathway, NF-κB inflammatory signaling, and lipogenic regulators SREBP-1, C/EBPβ, and PPARγ. Savas et al. (Lancet Diabetes Endocrinol, 2026; PMID 42208956) is a clinical review synthesizing evidence from randomized controlled trials and meta-analyses of approved and late-stage investigational obesity medications, including mazdutide alongside GLP-1 receptor agonists and multi-agonist agents.
