Description
MK-677 (ibutamoren) is a synthetic small molecule that binds the growth hormone secretagogue receptor (GHS-R1a), the same receptor activated by the endogenous peptide hormone ghrelin. It has been studied in animal models for its downstream activity along the hypothalamic-pituitary somatotropic axis and for GHS-R1a-linked signaling in kidney and brain tissue. The way a spare key opens a lock built for a different original key, MK-677 is designed to activate the ghrelin receptor through an orally available small molecule rather than the native peptide.
Lee et al. (Yonsei Medical Journal, 2018; PMID 30450851) studied oral MK-677 in rats over a six-week period, observing a transient rise in peak growth hormone concentration alongside an increase in hypothalamic somatostatin expression, without a corresponding change in body growth or IGF-1 levels at the study's endpoint. Sales da Silva et al. (Molecular and Cellular Endocrinology, 2020; PMID 32814069) used MK-677 as a GHS-R1a agonist tool compound in Wistar and spontaneously hypertensive rats to examine receptor-linked renal function and hemodynamic parameters. Tian et al. (Journal of Alzheimer's Disease, 2019; PMID 31594237) examined MK-677 in a transgenic mouse model of amyloid-beta pathology, finding increased hippocampal neurogenesis markers without a corresponding change in amyloid deposition, synaptic loss, or microglial activation measures.
