Description
HHB combines seven B-vitamin cofactors — biotin, niacinamide, niacin, pantothenic acid, thiamine HCl, choline, and inositol — each investigated in the biochemical literature for interaction with a distinct enzymatic step in keratinocyte metabolism or stratum corneum lipid production. The way a toolkit holds several distinct tools for different steps of one job, each B-vitamin component here is studied for a separate cofactor role in skin and nail-plate keratin biology, rather than as a single unified mechanism.
Tanno et al. (2000, British Journal of Dermatology) reported that nicotinamide (niacinamide) increased ceramide, free-fatty-acid, and cholesterol biosynthesis in cultured human keratinocytes in a concentration-dependent manner, and that topical application increased stratum corneum ceramide levels in an in vivo skin-barrier model. Cashman and Sloan (2010, Clinics in Dermatology) reviewed the keratin composition of the nail plate and catalogued nutritional deficiencies, including biotin deficiency, documented to affect nail-plate growth and structure. Patel, Swink, and Castelo-Soccio (2017, Skin Appendage Disorders) systematically reviewed published case reports of biotin supplementation for hair and nail changes, finding reported clinical improvement concentrated in cases involving an underlying biotin-deficiency pathology, while noting the evidence base for supplementation outside of deficiency states remains limited.
