Adamax (Ac-MEHFPGPAG-NH2) is a synthetic octapeptide derivative of Semax, a well-characterized ACTH(4-10) fragment analog. The molecule incorporates an adamantane cage substitution and terminal modifications (N-terminal acetylation, C-terminal amidation) drawn from the Peptide 021 design, giving it a distinct structural profile from the parent heptapeptide. The adamantane group is a lipophilic cage structure widely used in research chemistry to modify the physicochemical behavior of parent molecules under study.
The parent compound Semax has been studied across cellular and rodent models for its interactions with neurotrophin signaling pathways. Dolotov et al. (2006) examined Semax binding activity and brain-derived neurotrophic factor (BDNF) protein levels in the rat basal forebrain. Shadrina et al. (2010) compared NGF and BDNF gene-expression dynamics in a Wistar rat model of Semax exposure. Agapova et al. (2008) reviewed Semax effects on BDNF-related gene expression in rodent neural tissue. Adamax has not itself been characterized in the primary neurobiology literature; its structural design suggests laboratory investigation of how adamantane substitution alters the behavior of the Semax scaffold in comparative research models.
